中文摘要: 研究表明创伤后应激障碍(PTSD)是一种脑功能网络异常的精神障碍。患者脑功能系统间复杂的交互作用伴随局部神经环路(内侧前额叶-海马-杏仁核)的功能失调,是PTSD的神经环路理论模型之一,但目前对PTSD脑网络连接模式的认识尚不充分,神经影像学研究结果存在较大差异。针对以上现状,本项目拟开展如下工作:(1)探明前额叶-海马-杏仁核环路的功能失调是否为PTSD特异性基础假说,对三个关键节点的连接关系及其因果效应进行分析,构建PTSD局部神经有向网络。(2)探寻静息态下PTSD自发脑活动模式以及脑内在固有网络相对非PTSD患者的异常模式。(3)基于图论的复杂网络方法构建结构和功能大尺度网络,提供PTSD多模态脑网络连接神经影像表征。本研究可探明PTSD认知损伤及功能评价相关脑区的特定网络连接模式,从网络层面尝试为PTSD的病理生理机制及早期识别提供影像学靶标。

英文摘要: Evidence from studies suggests that post-traumatic stress disorder (PTSD), as a mental disorder, is believed to manifest from dysfunction in the functional network. Complex integrations in functional systems accompanied with the dysregulation of regional neurocircuit (medial prefrontal cortex-hippocampus-amygdala) are thought to be one of the theoretical models of PTSD. But we currently know very little about the patterns of brain functional connectivity in PTSD, and the neuroimaging findings are quite different. In line with the current situation, the current project aims to: (1) investigate the specificity of the medial prefrontal cortex-hippocampus-amygdala dysregulation model to PTSD versus non-PTSD, model the relationship within the three nodes, and build the regional network of effective connectivity; (2) detect the abnormal spontaneous brain activity and the disruption of intrinsic network in PTSD during resting state; (3) explore the large-scale structural and functional brain networks based on the graph theory, and detect the characterization of the brain connectivity networks in PTSD using multi-modal neuroimaging techniques.
