中文摘要: 小花棘豆是新疆分布危害最严重的毒草,目前国内外对小花棘豆中毒的症状研究较多,但鲜有小花棘豆中毒对动物抗氧化机能及细胞损伤的研究,其作用机理尚不清楚。本研究拟建立和田羊小花棘豆中毒模型,通过检测中毒动物机体自由基水平、抗氧化酶活性及基因表达量,分析小花棘豆中毒对和田羊脂质抗氧化作用及损伤、蛋白质和DNA的氧化损伤、细胞外基质的氧化损伤作用;然后提取小花棘豆主要毒性成分苦马豆素,通过体外细胞培养技术,应用DPH为探针采用荧光偏振测定苦马豆素对和田羊细胞膜结构与功能的影响,探讨小花棘豆中毒对和田羊细胞抗氧化相关信号分子激活程度的影响,全面分析小花棘豆中毒对和田羊抗氧化机能的影响,为和田羊小花棘豆中毒的防治提供新的思路。

英文摘要: Oxytropis glabra DC has extremely wide distribution and most seriously poisonous in Xinjiang Hui Autonomous Region of China. At home and abroad,there is not enough research on the Anti-Oxidant ability and cell damage on animals by poisoning of O.glabra,the mechanism of toxicity on O.glabra is not very clearunderstanding, making the control technology of O.glabra poisoning disease is slowly developed. This study is intended on the basis of previous research, establishment of Hetian sheep poisoning by O.glabra to detect serous, radical content, and hepatic redox status and the mRNA expression of antioxidant enzymes, for further research analyze the effects of lipid anti-oxidation, oxidation damage of protein, DNA and substruction besides cell; then extract swainsonine from O.glabra, creat the model of vitro cortical neurons cell and identification of its activity, investigate the protective effect of swainsonine on antioxidantive defense system and lipid peroxidation with DPH. Investigate effects of different peroxides on the expression of antioxidant related signaling factors of Hetian sheep by poisoning of O.glabra, Comprehenslly analyze the influence of antioxidation function of Hetian sheep as poisoning by O.glabra, reveal the toxicity mechanism of swainsonine in future, provided new opinion of preventionism and therapy for Hetian sheep by poisoning of O.glabra.
