中文摘要: 甘露(聚)糖结合凝集素(Mannan-binding lectin,MBL)在机体免疫中起重要作用。项目组的前期研究表明,不同MBL基因型绵羊血浆中MBL蛋白表达水平不同,对MP的抗性也不同。在人类中已有利用重组MBL蛋白治疗支原体肺炎的案例,但到目前为止MBL蛋白抗肺炎支原体作用方式及机理仍然还不清楚,这就给开发利用绵羊MBL基因带来了不确定因素。本研究拟从MBL基因入手,以绵羊肺炎支原体为疾病模型,构建MBL真核表达载体,体外表达MBL蛋白,通过外源注入的方法,检测注入后具有不同抗性基因型绵羊体内相关抗体及免疫因子变化,同时利用绵羊支气管粘膜细胞或肺泡上皮细胞作为细胞模型,体外研究绵羊肺炎支原体侵染细胞的抗性机制;采用转录组测序技术和miRNA测序技术,分析MBL不同基因型引发的多基因表达谱变化;以上研究结果综合分析MBL作为一线抗感染因子的分子调控机制,为利用MBL基因提供依据。

英文摘要: The Mannan-binding lectin(MBL) plays an important role in body immune. The previous study indicated that MBL protein expression levels are always different within different genotypes,and the MP resistance is different. The recombinant MBL has been used in human beings ,but the mechanism of MBL protein on anti-mycoplasma pneumonia is unclear so far, it is uncertain to develop and utilize sheep MBL. In this study, based on MBL gene and pneumonia mycoplasma used as disease model,a eukaryotic expression system which expresses MBL will be constructed. Changes of antibodies and immune factors in sheep of different genotypes after exogenous injection of the MBL expression vector, artificial infection of the bronchial mucosa cells or alveolar epithelial cells with pneumonia mycoplasma will be measured. Cellular mechanisms of mycoplasma infection will be investigated. In order to look into gene expression changes caused by genotype difference of MBL, gene chip technology and miRNA sequencing will be used in this research. These results will be used to analysis the molecular regular mechanism of MBL as the primary resistant infectious factor. The study will establish a theoretical basis to explore and utilize MBL gene.
