中文摘要: 热应激会给畜禽养殖造成严重危害,导致生产性能降低,繁殖力下降,免疫力低下,传染病发生等,对于理想的抗热应激药物研究越来越受到重视。本项目开展了牛磺鹅去氧胆酸(TCDCA)的抗热应激作用及其机制方面的研究。主要内容包括:(1)建立了小鼠热应激免疫抑制模型,证实了TCDCA能够显著提高热应激小鼠的采食量和饮水量;可以使热应激小鼠皮质酮和甲状腺素T3分泌、肝组织HSP70 mRNA转录以及免疫功能的紊乱状态得到改善,趋于维持在正常水平。(2)发现了TCDCA可以显著降低热应激AA肉鸡死亡率和极显著增加热应激海兰褐仔鸡的体重。(3)为了模拟热应激对动物机体下丘脑-腺垂体-肾上腺轴的影响,利用地塞米松建立了小鼠不良反应模型。证实了TCDCA可以减轻地塞米松引起的小鼠肾上腺和胃肠道损伤、恢复小鼠免疫功能和对抗负重游泳应激等。(4)开展了TCDCA对小鼠免疫器官及巨噬细胞系RAW264.7细胞凋亡和增殖影响的研究,发现TCDCA对小鼠脾脏、胸腺、肠系膜淋巴结及RAW264.7细胞系具有抗凋亡和促增殖作用。以上研究证实了TCDCA具有抗热应激作用,为今后将TCDCA开发成为抗热应激药物奠定了基础。

英文摘要: Heat stress can cause serious harm to livestock and poultry cultivation,leading to decreased in performance,the decline in fecundity,low immunity,infectious diseases,so research about the ideal thermal stress drug has been paid more and more attention.The study about the effects and mechanisms of taurochenodeoxycholic acid ( TCDCA ) in the project. The main contents include:(1)The model of immune suppression of thermal stress in mouse was established, and confirmed that TCDCA can obviously raise the feed intake and water consumption in the thermal stress mice,improved the state of disorder including the secretion of corticosterone and thyroxine T3, mRNA transcriptional level of HSP70 in liver tissue and the immune function in the thermal stress mice.(2)TCDCA can significantly reduce the death rates of the heat stressed AA broiler and very significantly increase the body weights of the heat stressed Hy-Line chicks were discovered.(3)The adverse reaction model was established using dexamethasone in mice in order to simulate the influence of thermal stress to the animal organism hypothalamus - pituitary and adrenal axis. TCDCA can reduce the injury of the adrenal gland and the gastrointestinal tract, restore the immune function and resist to swimming stress by dexamethasone in mice.(4)The effects of TCDCA to the cell apoptosis and proliferation of the mice immune organ and macrophage cell line RAW264.7 were studied, it found that TCDCA had the effects in the anti-apoptosis and promoting proliferation to the murine spleen, thymus, mesenteric lymph node and RAW264.7 cell lines in mice. The above research confirmed that TCDCA had the effects of thermal stress, it were laying the foundation further that TCDCA was developed the medicines against heat stress in the futher.
